Abstract
Većinom sve vrste medicinskog, ljekovitog i aromatičnog bilja sadrže velike količine polisaharida, polifenola i ostalih sekundarnih metabolita. Cilj rada bio je opisati postupak izolacije molekule DNA te utvrditi kvalitetu i količinu izolirane DNA kod biljnih vrsta koje se koriste u ljekovite svrhe. Biljne vrste korištene u istraživanju su: ružmarin, kadulja, kamilica, lavanda, ljupčac, menta, ruta i anis. One su uzgajane u kontroliranim uvjetima i nisu bile pod utjecajem vanjske sredine. Prilikom izolacije korištene su tri metode: CTAB, SDS i FA-UniZG-DNA. Tekući dušik korišten je kod homogenzacije biljnog tkiva, dok je kvaliteta i količina izolirane DNA testirana elektroforetski. CTAB metoda se pokazala najuspješnijom za izolaciju DNA iz svih ispitivanih biljnih vrsta.
Abstract (english)
Most of all medical, medicinal and aromatic plants contain large amounts of polysaccharides, polyphenols and other secondary metabolites. Aim of this resarche was to describe the process of isolation of DNA molecules and to determine the quality and quantity of isolated DNA in plant species which are used in medicinal purposes. Plant species used in this research are: rosemary, sage, chamomile, lavender, lovage, mint, anise and routes. They were grown in controlled area and hadn't any impact of extrnal environment. We used used three types of isolation methods: CTAB, SDS and FA-UniZG-DNA. Liquid nitrogen was used in homogenization of plant tissue, while purity and quality of isolated DNA was estimated by electrophoresis.
