Abstract
U ovom istraživanju ispitana je genetska varijabilnost svojstava kvalitete germplazme soje i genetska udaljenost genotipova temeljem utvrđivanja mase 1000 zrna, kemijskih analiza zrna i molekularnih analiza. Ciljevi ovog istraživanja bili su na 22 genotipa soje: (1) procjeniti genetsku varijabilnost svojstava kvalitete zrna i genetsku udaljenost germplazme soje na temelju utvrđivanja mase 1000 zrna te kemijskih analiza zrna koje su uključivale određivanje koncentracije bjelančevina i ulja, sastava i koncentracije masnih kiselina i saharida te sastava i sadržaja izoflavona; (2) procjeniti genetsku varijabilnost svojstava kvalitete zrna i genetsku udaljenost genotipova soje uz pomoć SSR biljega izabranih temeljem rezultata studija povezanosti polimorfizma DNA biljega i pojedinih istraživanih svojstava kvalitete zrna; (3) odabrati genotipove s poželjnim svojstvima kvalitete zrna; (4) identificirati najudaljenije sorte s poželjnim svojstvima kvalitete kao potencijalne, nove roditeljske kombinacije za buduće programe križanja. Tijekom tri vegetacijske sezone (2010. – 2012.), na Poljoprivrednom institutu Osijek postavljen je poljski pokus u dva ponavljanja po slučajnom bloknom rasporedu. Svake godine pokusa nakon žetve određena je masa 1000 zrna te su napravljene kemijske analize kojima se utvrdila koncentracija bjelančevina i ulja, sastav i koncentracija masnih kiselina i saharida te sastav i sadržaj izoflavona u zrnu soje. Na kraju trogodišnjeg pokusa napravljene su i molekularne analize. Statistička obrada rezultata ovih analiza uključivala je izračun standardnih mjera varijacije, analizu varijance, utvrđivanje udaljenosti genotipova temeljem standardiziranih srednjih vrijednosti svojstava kvalitete zrna te izradu pripadajućeg UPGMA dendrograma, utvrđivanje genetske udaljenosti po Nei-u (1973.) temeljem molekularnih analiza uz pomoć 10 mikrosatelitnih biljega te izradu pripadajućeg UPGMA dendrograma, analizu molekularne varijance za genotipove porijeklom sa Poljoprivrednog instituta Osijek te usporedbu podataka različitih vrsta analiza Mantelovim testom. Nakon završene analize podataka utvrđeno je da unutar istraživanog seta biljnog materijala postoji genetska raznolikost u svojstvima kvalitete zrna što je i potvrđeno na molekularnoj razini.
Abstract (english)
This research investigated the variability of soybean germplasm seed quality by determining 1000 seed weight, chemical analyses of seed and molecular analysis of 22 soybean genotypes. Aims of this research were: (1) assessing genetic variability of seed quality and genotypes' genetic distance on the basis of measuring 1000 seed weight and by determining soybean seed protein and oil concentrations, composition and concentrations of fatty acids and saccharides as well as isoflavone composition and content; (2) assessing genetic diversity and genetic distance of genotypes with the help of SSR markers chosen on the basis of relevant association studies between used markers and investigated seed quality parameters; (3) choosing genotypes with favourable seed quality; (4) among genotypes with favourable seed quality, choosing those genetically most distant for future crossing programmes. Field trials were set up in randomized complete block design with two replications, at the Agricultural Institute Osijek during three growing seasons (2010-2012). Chemical analyses and determining 1000 seed weight were performed each year after harvest while molecular analysis was done at the end of the three year trial. Statistical analyses of the results included: calculating basic measures of variation, analysis of variance, determining genotypes' genetic distance with standardised average values of seed quality parameters and contructing corresponding UPGMA dendrogram, determining genetic distance according to Nei (1973) on the basis of molecular analysis with 10 microsatellite markers as well as constructing corresponding UPGMA dendrogram, analysis of molecular variance for Agricultural Institute Osijek's genotypes and comparing the results of different types of analyses with Mantel's test. Analysed data showed exsistance of genetic diversity in seed quality among tested plant materials, wich was confirmed on the molecular level also.
