Objectives: The aim of this study is to determine the effect of lipid enriched medium on the morphology of the cell line SH-SY5Y human neuroblastoma, also to determine the effect of simvastatine on the morphology of cells grown in standard or in lipid enriched medium and to isolate lipid rafts and to determine their composition using the Western blot method. Study design: Matched pair study. Material and methods: This research has been conducted by using the SH-SY5Y cell line of human neuroblastoma. The cells were then treated with a standard medium, sivastatine, lipid enriched medium, lipid enriched medium with the addition of simvastatine, DMSO, DMSO with the addition of simvastatine. After 24 hours of treating, the morphology of cells prepared this way has presented itself with a immunocytochemical staining onto the markers of cytoskeletons SMI31, SMI32, SMI312. The results were analyzed with ImageJ computer program. The isolation of lipid rafts has been done by ultracentrifugation. Using the Western blot method, the composition of lipid rafts was determined with the help of raft and non-raft markers, as well as the antibodies on the AMPA receptor as a neuroplasticity marker. Results: Staining with the Cholera toxin shows that cells treated with simvastatine have the longest length of neurites compared to control group and cells treated with fatty acids have the shortes length of neurites. The quantification of immunocytochemical staining on SMI312 displays statistically significant differences between cells treated with simvastatine in comparison to the control group, simvastatine and fatty acids, simvastatine and fatty acids combined with simvastatine, furthermore fatty acids combined with simvastatine display a significant difference compared to cells treated with DMSO. Statistically there is an important difference between cells treated with fatty acids compared to the control group and DMSO at cells stained on SMI32. Staining on SMI31 did not show any statistically important differences. Conclusion: Simvastatine encourages SH-SY5Y cell line neuritogenesis.
