Abstract
CILJEVI ISTRAŽIVANJA. Ciljevi ovog istraživanja bili su uspoređivanje površine presjeka glomerula, kao i uspoređivanje promjera kanalića, visine epitela i promjera lumena kanalića kod miševa divljeg tipa i miševa s isključenim genom Tff3 te ispitivanje postojanja razlika u navedenim parametrima između tih dviju skupina miševa. USTROJ STUDIJE. Istraživanje je provedeno kao presječna studija u kojoj su korišteni uzorci mišjih bubrega iz arhive Zavoda za histologiju i embriologiju. MATERIJAL I METODE. Za istraživanje su korišteni bubrezi miševa divljeg tipa te miševa s isključenim Tff3 genom. Korišteni su uzorci pet životinja po skupini (ukupno deset miševa). Preparati su obojeni metodom PAS (Perjodna kiselina i Schiff-ov reagens), nakon čega su analizirani na osnovi histomorfološke građe i parametara. Mjerenja parametara izvršena su pomoću programa FIJI na pet fotografija kore i trima fotografijama srži za pojedinu životinju. REZULTATI. Miševi s isključenim Tff3 genom imali su veći promjer proksimalnog kanalića i širinu njegova lumena nego miševi divljeg tipa. Analizom ostalih vrijednosti nije uočena značajna razlika mjerenih vrijednosti u kori i srži bubrega. ZAKLJUČAK. Postoje značajne razlike u promjeru proksimalnih kanalića te širini lumena proksimalnih kanalića kod miševa divljeg tipa i miševa s isključenim genom Tff3, za razliku od površine presjeka glomerula, promjera distalnih i sabirnih kanalića te visine epitela i širine lumena proksimalnih, distalnih te sabirnih kanalića.
Abstract (english)
OBJECTIVES: The objectives of this study were to compare the cross-sectional area of the glomeruli as well to compare tubular diameter, epithelial height and tubular lumen diameter in wild-type mice and Tff3 knock-out mice and to examine whether there are differences in these parameters between the two groups of mice. STUDY DESIGN: The study was done as a cross-sectional study using samples of mice kidneys from the archives of the Department for Histology and Embryology. MATERIAL AND METHODS: Five samples of mice kidneys of wild type and five samples of Tff3 knock-out mice were used for this research (ten mice in total). The kidney slides were stained by PAS (Periodic Acid Schiff reaction) after which they were analyzed on the basis of histomorphological structure and parameters. Parameter measurements were performed using the FIJI program on five photographs of cortex and three photographs of the marrow for each animal. RESULTS: Tff3 knock-out mice had a larger proximal tubule diameter and lumen width than wild-type mice. The analysis of other values did not show a significant difference between the measured values in the cortex and marrow of the kidney. CONCLUSION: There are significant differences in the diameter of the proximal tubules and the width of the lumen of the proximal tubules in wild-type mice and Tff3 knock-out mice, unlike to the glomerular cross-sectional area, distal and collecting duct diameter, epithelial height and lumen width of proximal, distal and collecting ducts.
