Abstract
UVOD: Rak je bolest koju karakteriziraju nekontrolirana stanična proliferacija i sposobnost širenja na druge stanice i tkiva. Primjena ferocena aktivno je područje istraživanja, a izvješća pokazuju in vitro i in vivo aktivnosti protiv različitih bolesti, a područje najvećega interesa su maligne bolesti. CILJ: Ispitati utjecaj novih derivata ferocena na rast leukemijskih stanica u uvjetima in vitro. Ustanoviti povezanost između strukture i koncentracije novih spojeva i inhibicije rasta. MATERIJAL I METODE: Korištene su četiri stanične leukemijske linije u triplikatu: K562, CCRF-CEM, HL60 i THP1 koje su tretirane u koncentracijama 1×10-4 M, 1×10-5 M i 1×10-6 M novo-sintetiziranih derivata ferocena. Antiproliferativna aktivnost određena je MTT testom nakon izlaganja stanica derivatima ferocena kroz 72 sata. REZULTATI: Najznačajniji citotoksični učinak derivati ferocena na svim staničnim linijama ostvaruju pri apliciranoj koncentraciji od 1×10-4 M, a najmanji pri koncentraciji od 1×10-6 M. ZAKLJUČAK: Novi derivati ferocena sposobni su inhibirati rast leukemijskih staničnih linija. Ostaje za istražiti mehanizam djelovanja ispitivanih novih derivata ferocena.
Abstract (english)
INTRODUCTION: Cancer is a disease characterized by uncontrolled cell proliferation and the ability to spread to other cells and tissues. The application of ferrocene is an active area of research, and reports show in vitro and in vivo activities against various diseases, while the area of greatest interest is malignant diseases. OBJECTIVE: To investigate the effect of new ferrocene derivatives on the growth of leukemic cells in vitro. To establish an association between the structure and concentration of new compounds and growth inhibition. MATERIAL AND METHODS: Four cell leukaemia lines in triplicate were used as listed: K562, CCRF-CEM, HL60 and THP1 treated at concentrations of 1×10-4 M, 1×10-5 M and 1×10-6 M newly synthesized ferrocene derivatives. Antiproliferative activity was determined by MTT assay after exposure of cells to ferrocene derivatives for 72 hours. RESULTS: The most significant cytotoxic effect of ferrocene derivatives on all cell lines is achieved at an applied concentration of 1×10-4 M and the smallest at a concentration of 1×10-6 M. CONCLUSION: New ferrocene derivatives appear to be able to inhibit the growth of leukemic cell lines. It remains to investigate the mechanism of action of the investigated new ferrocene derivatives.
