Abstract
Mikronukleus (MN) test na limfocitima periferne krvi jedna je od najvaţnijih metoda koje se primjenjuju u biološkom nadzoru, a zasniva se na na 72-satnome uzgoju limfocita periferne krvi u hranjivom mediju te izradi mikroskopskih preparata koji se pregledavaju pod svjetlosnim mikroskopom. Cilj ovog istraţivanja je ispitati genotoksični učinak ksilola, metanola i ledene octene kiseline i odrediti indeks proliferacije limfocita periferne krvi zaustavljanjem citokineze pri različitim koncentracijama kemikalija mikronukleus testom. Analizirani su limfociti triju ispitanika tretirani s tri različite koncentracije svake od navedenih kemikalija. Biljeţen je ukupan broj MN, ukupni broj stanica s MN, raspodjela MN u stanicama, broj nukleoplazmatskih mostova (NPB) i nuklearnih pupova (Nbud) te indeks proliferacije stanica. Ksilol, metanol i ledena octena kiselina uzrokuju statistički značajno povećani broj mikronukleusa i smanjenje indeksa proliferacije (t test, P<0,05). Pokazano je da je mikronukleus test vrlo dobar pokazatelj genotoksičnog i citotoksičnog djelovanja ispitivanih kemikalija.

The micronucleus (MN) test on lymphocytes of peripheral blood is one of most important methods used in biologic monitoring. It is based on the 72-hour cultivation of lymphocytes of peripheral blood in nutrient medium and making microscopic preparations which are observed under optical microscope. The aim of this research was to examine the genotoxic effect of xylene, methanol and glacial acetic acid, and to determine the proliferation index of peripheral blood lymphocytes by cytokinesis at different chemical concentrations in the micronucleus test. The lymphocytes of three participants are analyzed with three different concentrations of each of the enlisted chemicals. The total number of MN, the total number cells with MN, the distribution of MN in cells, the number of nucleoplasmatic bridges (NPB) and nuclear buds (Nbud) and cell proliferation index were recorded. Xylene, methanol and glacial acetic acid cause a significantly increased number of micronuclei and lowered proliferation index (t test, P<0,05). It was proven that micronucleus test is a very good indicator of genotoxic and cytotoxic action of the tested chemicals.
