Abstract
Cilj istraživanja: Napraviti morfometrijski prikaz jetre i slezene kod odraslih miševa divljeg tipa, miševa s isključenim genima B4Galnt1 i miševa hranjenih kuprizonom. Nacrt studije: Istraživanje je provedeno kao studija parova. Materijali i metode: Za provedeno istraživanje se koristilo tkivo jetre i slezene miševa divljeg tipa (WT), WT miševi hranjeni kuprizonom i genetički izmijenjenih miševa B4Galnt1 (KO). Tkiva jetre i slezene su bojana klasičnim histološkim bojanjima te su preparati pogledani pod mikroskopom i slikani. Dobiveni rezultati su se obrađivali u računalnim programima Fiji i Statistica 12. Rezultati: Kod miševa s isključenim genom B4Galnt1 i miševa hranjenjih kuprizonom dolazi do smanjenja ukupnog broja masnih kapljica u jetri. Količina željeza u jetri životinja hranjenih kuprizonom je statistički značajno manja u odnosu na divlji tip i KO B4Galnt1 mišji model. Nema morfoloških promjena slezene kod sva tri modela miša. Zaključak: Kod miševa hranjenih kuprizonom i KO miševa dolazi do promjena u jetri, no promjene u slezeni nisu zapažene.
Abstract (english)
Aims: To do morphometric survey of liver and spleen in wild type adult mice, B4Galnt1 knockouts and cuprizone - fed mice. Study scheme: A case- control study. Materials and methods: Liver and spleen tissue of wild type mice (WT), cuprizone - fed wild type mice and genetically modified mice (KO) were included in the conducted research. Liver and spleen tissue samples were stained with the classical histological stain dye, the tissue samples were examined under a light microscope and photographed by camera. The obrained results were processed in Fiji and Statistica 12 computer programmes. Results: In KO B4Galnt1 mice and cuprizone – fed mice, the total number of fat drops in the liver is reduced. The amount of iron in the liver of cuprizone-fed animals has statistically been significantly lower compared to the wild type and the KO B4Galnt1 mouse model. There are no morphological changes in the spleen of all three mouse models. Conclusion: There have been changes taking place in the liver of the cuprizone - fed and KO B4Galnt1 mice. There have not been any noticed spleen changes though.
