Abstract
Gangliozidi su posebna skupina glikosfingolipida koji sadrže jednu ili više sijalinskih kiselina. Njihova uloga uglavnom je povezana s prijenosom signala u lipidnim splavima staničnih membrana i neurorazvojem. Nalaze se u živčanom sustavu svih kralježnjaka, a jako mali broj studija odnosi se na analizu njihove prisutnosti u gujavicama. S obzirom na dostupne izvore, ovaj rad prvo je istraživanje izražajnosti gangliozida kod gujavica pomoću imunohistokemijske metode. Unatoč primjeni visokospecifičnih monoklonalnih protutijela IgG za glavne moždane gangliozide GM1,GD1a,GD1b i GT1b, imunohistokemijska metoda nije otkrila prisutnost gangliozida kod gujavice E.andrei. Modificirana metoda ekstrakcije i tankoslojne kromatografije (TLC) u konačnici je dala pozitivan rezultat, čime je ustanovljena prisutnost gangliozida s većim brojem sijalinske kiseline i GD1a u stražnjem dijelu tijela gujavice. Potrebna su dodatna istraživanja kako bi se otkrila adekvatna metoda koja bi postala standard u istraživanju gangliozida kod ovih organizama.
Abstract (english)
Gangliosides are a special group of glycosphingolipids that contain one or more sialic acids. Their role is mainly associated with signal transmission in lipid rafts of cell membranes and neurodevelopment. They are found in the nervous system of all vertebrates, and a very small number of studies have also reported their presence in earthworms. According to available sources, this is the first study of ganglioside expression in earthworms using the immunohistochemical method. Despite the use of highly specific monoclonal antibodies for the major brain gangliosides GM1, GD1a, GD1b and GT1b the immunohistochemical method did not detect the presence of gangliosides in earthworm E. andrei. However, modified extraction method and thin-layer chromatography (TLC) gave a positive result, identifying the presence of gangliosides with a higher number of sialic acids and GD1a in the posterior part of the earthworm's body. Further research is needed to discover an adequate method that could become the standard for studying gangliosides in these organisms.
