Abstract
Sinaptička plastičnost je sposobnost centralnog živčanog sustava da se mijenja u odgovoru na promjene vanjskih uvijeta. Jedan od oblika sinaptičke plastičnosti, dugoročna potencijacija (LTP), događa se u neuronima kao odgovor na promijene u učestalosti i intezitetu podražavanja određenih sinapsi. Rezultat dugoročne potencijacije očituje se u jačanju tih sinapsi, odnosno stvaranju čvrstih veza između membrana dvaju susjednih neurona. Jedna od molekula koja sudjeluje u stvaranju veza između... More sinapsi je NCAM (engl. Neural Cell Adhesion Molecule), transmembranski protein iz superporodice imunoglobulina koji homofilnim interakcijama dovodi do stanične adhezije i na taj način sudjeluje u stvaranju veza na sinapsama, te pokreče unutarstanične kaskade reakcija koje pomažu u održavanju ovih veza. Polisijalinskom kiselinom modificirani oblik ove molekule (PSA-NCAM) potiče plastičnost na način da fizički ometa stvaranje homofilnih interakcija te omogučuje migraciju neurona. PSANCAM najobilnija je u razvojnim stadijima centralnog živčanog sustava dok je u odraslom mozgu u najvećim koncentracijama prisutna u regijama koja zadržavaju sposobnost plastičnosti. Less
Abstract (english)
Synaptic plasticity is the ability of the central nervous system to adapt to various changes in the enviroment. One of the forms of synaptic plasticity, long term potentiation (LTP), occurs in neurons as a response to changes in frequency and intensity of stimulus on certain synapses. The end result of long term potentiation is the strenghtening of syapses, respectively forming tight connections between membranes of opposing neurons. More connectiones between synapses is NCAM ( Neural Cell Adhesion Moleucle), a transmembrane protein of the immunoglobuline, it forms homophilic interactions which lead to adhesion of cell walls on the synapses, also it causes an intrecellular cascade of biochemical reactions inside the cell that help preserve the connections. A polysialated form of NCAM (PSA-NCAM) promotes synaptic plasticity by physicly interfering with the formation of homophilic interactions, in this way it enables the migration of neurons. PSA-NCAM has been mainly found in the developing brain, in the adult brain it is most abundant in regions that remain plastic. Less
