Abstract
Gangliozidi, kiseli glikosfingolipidi, lipidni su dijelovi stanične membrane koji su poznati kao modulatori transmembranskog signaliziranja. Brojna istraživanja provedena su na ulozi gangliozida u inzulinskoj rezistenciji, patološkom stanju kojeg karakterizira izostanak odgovora stanice ili tkiva na fiziološko lučenje inzulina. Dokazano je kako gangliozidi mogu uzrokovati inzulinsku rezistenciju neposredno, ali i posredno preko citokina TNFα. Neposredna rezistencija kao posljedica povećane... More ekspresije GM3, najjednostavnijeg gangliozida, podrazumjeva inhibiciju autofosforilacije inzulinskog receptora uslijed čega dolazi do prekida signalnog puta. Posredni utjecaj gangliozida na rezistenciju uključuje djelovanje TNFα, citokina imunosnoga sustava, koji povećava koncentraciju GM3 na staničnoj membrani adipocita. U ovom radu navedeni su mehanizmi i istraživanja koja podupiru navedene primjere i ističu važan utjecaj GM3 kao i TNFα u inzulinskoj rezistenciji, odnosno patologiji šećerne bolesti tipa 2. Less
Abstract (english)
Gangliosides, acid glycosphingolipids, are lipid components of the cell membrane known as transmembrane signal modulators. Numerous studies have been conducted on the role of gangliosides in insulin resistance, a pathological condition characterized by the absence of the cell or tissue response to physiological secretion of insulin. It has been shown how gangliosides can cause insulin resistance directly, but also indirectly through the cytokine of TNFα. Immediate resistence as a result... More of increased expression of GM3, the simplest ganglioside, implies the inhibition of insulin receptor autophosphorylation resulting in the signal path interruption. The mediating effect of gangliosides on resistence involves the action of TNFα, a cytokine immune system, which increases the concentration of GM3 on the adipocyte cell membrane. In this paper, the mechanisms and researches that support these examples are listed and they highlight the important influence of GM3 as well as TNFα in insulin resistance, or diabetes mellitus type 2.
