Abstract
Gljiva Cryphonectria parasitica uzročnik je raka kore europskog pitomog kestena. Osim na kestenu ovaj patogen čini štetu i na drugim vrstama iz porodice Fagaceae, ali u znatno manjoj mjeri. Srećom, u Europi je primijećen prirodni oporavak stabala kestena zahvaljujući pojavi hipovirusa koji smanjuje njezinu virulentnost i sposobnost sporulacije. Širenje hipovirusa u populaciji gljive dovodi do oporavka kestena te uspješne biološke kontrole. Međutim, prijenos virusa ograničen je vegetativnom nekompatibilnošću (vic) gljive, pri čemu se virus brzo širi između jedinki gljive istog VC tipa, a sporoili se uopće ne širi između jedinki različitog VC tipa. Cilj ovog istraživanja bio je karakterizirati uzorke gljive C. parasitica na osnovi morfologije, tipa vegetativne kompatibilnosti (VC) i molekularnihanaliza. Rezultati morfologije pokazali su prisutnost 77% žutih virulentnih sojeva i 23% bijelih sojeva gljive zaraženih hipovirusom. Najzastupljeniji VC tip gljive bio je EU 1 (69%) zatim EU 2 (23%) te EU12 sa 8%. Hipovirulentnim sojevima gljive dokazana je prisutnost hipovirusa CHV1 izolacijom hipovirusne dsRNA i RT-PCR metodom, dok je analiza polimorfizma duljine restrikcijskih odsječaka(RFLP) potvrdila da CHV1 pripada talijanskom podtipu, CHV1 I.

Cryphonectria parasitica is the causal agent of chestnut blight. Besides to chestnut, this pathogen causesdamage to other species of Fagaceae family, but to a much lesser extent. Fortunately, a natural recoveryof chestnut trees has been observed due to appearance of hypovirus which infects fungus, and reducesits virulence, sexual reproduction and sporulation. Spreading of hypovirus in fungus population leads tochestnut recovery and successful biological control. However, transmission of the hypovirus is limitedby the vegetative incompatibility (vic) system. Hypovirus spreads quickly between fungal individuals ofthe same VC type and slowly or it does not spread at all between the individuals with different VCtypes. Isolates of the fungus C. parasitica were characterized based on morphology, type of vegetative compatibility and RT-PCR/RFLP analysis. Morphology results showed 77% yellow and 23% whitehypovirulent strains. Three VC types were identified, with the dominant VC type EU1 (69%) followedby the EU2 (23%) and EU12 (8%). The presence of hypovirus CHV1 in all tested hypovirulent strainswas confirmed by dsRNA isolation and RT-PCR method, while the analysis of restriction fragmentlength polymorphism (RFLP) confirmed that CHV1 isolates belong to Italian subtype, CHV1 I.
